Responsibilities
Position Purpose:
Develop and implement chemical biology techniques to advance projects across the Oncology portfolio, primarily through the design and application of chemical probes in complex biological systems.

Primary Duties:
- Design and carry out experiments with functional chemical probes to address specific project needs across the Oncology Research Unit (ORU).
- Work in collaboration with ORU biology to design and execute experiments aimed at answering key biological questions.
- Work in collaboration with the Worldwide Medicinal Chemistry Design and Synthesis groups to conceive and prepare chemical probes.
- Apply and/or develop affinity/activity-based proteomic probes to profile the functional state of enzyme targets in native biological systems and assess selectivity and/or safety of compounds.
- Design of photoaffinity probes and conduct photoaffinity labeling experiments for covalent capture of targets.
- Build effective partnerships with the biophysics group in the ORU and the MS proteomics group in Worldwide Medicinal Chemistry.
- Expand the global chemical proteomics toolbox. Where applicable, use click chemistries to build functional molecules in situ.
- Use chemical proteomics to elucidate biological targets of active molecules from phenotypic screens to expedite drug discovery.
- Design and application of fluorescent probes for live cell imaging studies.

Qualifications
Educational Qualifications:
Doctoral level degree or equivalent experience in chemical biology or related area

Training and Experience:
- Strong track record of delivering biological insights through chemical biology experimentation.
- Highly skilled synthetic chemist with comprehensive knowledge of building chemical biology probes.
- Highly skilled laboratory experimentalist with experience of working on biological systems (cells/cell lysates/ex vivo tissues etc).
- Experience with standard methods used in chemical biology including: protein purification/enrichment, chromatography (size exclusion, ion exchange, affinity) immunoprecipitation, gel electrophoresis; Western blotting.

Preferred:
- Post doctorate in chemical biology or related area.
- Target identification and mechanism of action elucidation experience.
- Comfortable interpreting MS data for identification of target proteins.
- Experience working in the following areas would be desirable: Click chemistry; photoaffinity cross-linking; in-gel fluorescence; chemical modification of proteins; protein engineering; mammalian tissue culture, SILAC methodology; siRNA for target validation.
